A <a href=\"https:\/\/player.vimeo.com\/video\/434433967\" target=\"_blank\" rel=\"noreferrer noopener\">short video<\/a> describes a recent piece of work of the team relevant to the project.<\/p>\r\n\r\n\r\n\r\n<h5 class=\"wp-block-heading\"><strong>Why ascidians?<\/strong><\/h5>\r\n\r\n\r\n\r\n<p>Ascidians are a group of marine invertebrates. Their embryonic cell lineages and early embryonic stage morphologies have remained essentially identical since the group&#8217;s emergence about 400 million years ago. This suggests that they are subject to very strong developmental or evolutionary constraints (Lemaire et al. 2011). Ascidian embryogenesis is also very robust to environmental perturbations of temperature and salinity. The extreme evolutionary and environmental robustness of embryonic geometries and cell lineages contrasts with a rapid genetic divergence between species and intra- and inter-specific variability in gene expression.<\/p>\r\n\r\n\r\n\r\n<h5 class=\"wp-block-heading\"><strong>What is the project?<\/strong><\/h5>\r\n\r\n\r\n\r\n<p>The proposed experimental project will <strong>provide a quantitative assessment of the developmental robustness of a critical morphogenetic process, ascidian gastrulation<\/strong> (1, 3, 5), to two key environmental parameters (water temperature and salinity) and to genetic perturbations of the morphogenetic driving force apparatus (myosin II, Rho kinase, &#8230;). The project will study the magnitude of environmental or genetic variations compatible with the production of a viable larva. It will seek to identify the least &#8211; and most &#8211; robust developmental processes and time points, i. e. those that collapse first, or resist best to the perturbations. Finally, it will characterize the structure of the natural and experimentally-induced variability in the geometry and mechanical properties of embryos. This may lead to the identification of developmental modules.<\/p>\r\n\r\n\r\n\r\n<p>The project will involve <strong>advanced light-sheet imaging of live micro-injected embryos <\/strong>of the ascidian <em>Phallusia mammillata<\/em>, followed by the<strong> computational and statistical analysis<\/strong> of the acquired developmental movies (see 2, 4).<\/p>\r\n\r\n\r\n\r\n<h5 class=\"wp-block-heading\"><strong>Who funds the project, and how to apply?<\/strong><\/h5>\r\n\r\n\r\n\r\n<p>The project is funded by an ANR-NSF binational project grant and will be conducted in collaboration with 3 other teams: Prof. <a href=\"http:\/\/www.msc.univ-paris-diderot.fr\/~atef_asnacios\/index_en.html\" target=\"_blank\" rel=\"noreferrer noopener\">Atef Asnacios<\/a> (MSC, Paris), Prof. <a href=\"https:\/\/munrolab.uchicago.edu\/\" target=\"_blank\" rel=\"noreferrer noopener\">Edwin Munro<\/a> (U. Chicago, USA) and Prof. <a href=\"https:\/\/www.madhavmani.com\/\" target=\"_blank\" rel=\"noreferrer noopener\">Madhav Mani<\/a> (Northwestern University, Evanston, USA).<\/p>\r\n\r\n\r\n\r\n<p>Expected candidates will have a <strong>PhD in cell and developmental biology<\/strong>, an excellent track record of publications and oral communications, strong skills in fluorescent live imaging and some experience in the computational analysis of large datasets. Our overall goal is to bridge the molecular and cellular scales of analysis in order to provide an integrated view of a developmental program in time and space and to study its evolution. We first made use of the stereotyped and highly conserved cell lineage of ascidian embryos to decipher in detail their early transcriptional developmental program (Cell, 2003; Development 2006, 2007; Current Biology 2010). To understand how the lineage and morphogenetic program could be so well conserved in spite of extensive genomic divergence between ascidian species, we studied the architecture of ascidian cis-regulatory sequences and showed that is was flexible and robust to extensive TF binding site turnover (Development, 2005; Current Biology 2010; bioRxiv 2016). Thanks to the transparency of ascidian embryos, we could image and quantitatively describe early ascidian morphogenesis with cellular resolution (Current Biology, 2006; Science, 2020), and use this description to provide a molecular and mechanical model for the invagination of the endoderm at the beginning of gastrulation (Current Biology, 2010; Development 2020). To efficiently integrate heterogeneous datasets ranging from gene expression data to quantitative geometric descriptions of cell and tissue morphologies, we developed an integrative model organism database (ANISEED, Genome Research, 2010, Nucleic Acid Research 2016, 2018), which is the main database in the tunicate field. Our overall goal is to bridge the molecular and cellular scales of analysis in order to provide an integrated view of a developmental program in time and space and to study its evolution. We first made use of the stereotyped and highly conserved cell lineage of ascidian embryos to decipher in detail their early transcriptional developmental program (Cell, 2003; Development 2006, 2007; Current Biology 2010). To understand how the lineage and morphogenetic program could be so well conserved in spite of extensive genomic divergence between ascidian species, we studied the architecture of ascidian cis-regulatory sequences and showed that is was flexible and robust to extensive TF binding site turnover (Development, 2005; Current Biology 2010; bioRxiv 2016). Thanks to the transparency of ascidian embryos, we could image and quantitatively describe early ascidian morphogenesis with cellular resolution (Current Biology, 2006; Science, 2020), and use this description to provide a molecular and mechanical model for the invagination of the endoderm at the beginning of gastrulation (Current Biology, 2010; Development 2020). To efficiently integrate heterogeneous datasets ranging from gene expression data to quantitative geometric descriptions of cell and tissue morphologies, we developed an integrative model organism database (ANISEED, Genome Research, 2010, Nucleic Acid Research 2016, 2018), which is the main database in the tunicate field.
